High-Fidelity Production of In Vitro Transcription Plasmids with Long Poly(A) Sequences

Preserve long poly(A) tails for optimal mRNA plasmid stability

Due to their versatility for protein expression, synthetic mRNA plasmids have become an ideal tool for gene therapies. A key component of an mRNA plasmid is the poly(A) tail, which determines the structural integrity of the molecule. While long poly(A) sequences have generally resulted in increased stability of mRNA products, poly(A) tails with greater than 100 bases can be susceptible to truncations during production, resulting in unstable mRNA products.

To address this challenge, Azenta Life Sciences has developed an efficient protocol using a proprietary E. coli strain and optimized culturing methods to ensure long poly(A) sequence stability of at least 140 bases in mRNA plasmids.
