Advancing Recombinant Adeno-Associated Virus Vector Integration Analysis through Next-Generation Sequencing Optimization
Recombinant adeno-associated viral (rAAV) vectors have seen widespread application as a gene therapy delivery platform in preclinical and clinical studies. rAAV integration analysis, however, presents several challenges, including the random nature of rAAV integration sites and low expected integration frequency. In this poster, we present next-generation sequencing (NGS)-based methodologies for both rAAV integration analysis, using hybrid-capture-based target enrichment sequencing (TES), and lentivirus integration analysis using linear amplification method PCR (LAM-PCR).
